NAD+ precursor supplements (NMN, NR): what human trials actually show so far
The cellular-aging science behind NAD+ decline is well established. The human-trial evidence for supplementing it is considerably earlier stage.
NAD+ (nicotinamide adenine dinucleotide) is a coenzyme essential to cellular energy metabolism and DNA repair, and its levels reliably decline with age — this part of the underlying biology is well established across animal and cellular research. The commercial interest is in precursor supplements, primarily NMN and NR, which the body can convert into NAD+, on the theory that supplementing precursors could restore youthful NAD+ levels and their associated benefits.
Human trial data on NMN and NR remains considerably earlier stage than the cellular science suggests to consumers — small trials have generally confirmed these supplements do raise blood NAD+ levels, which establishes the basic mechanism works in humans, but trials measuring actual functional or health outcomes (rather than just the biomarker) have been smaller, shorter, and less conclusive than the animal literature, which has shown more dramatic effects on lifespan and healthspan in mice specifically.
The gap between animal-model results and human evidence is a recurring pattern in longevity research generally, not unique to NAD+ precursors — it's a reasonable reason for cautious optimism about a promising research direction, while being clear that current human evidence doesn't yet support the strength of claims sometimes made in supplement marketing built primarily on the mouse data.
